@#The tolerance of microorganism under acid stress is of significant importance to the growth and industrial production of bacterial strain. When the bacterial cells are exposed to the external acid environment, proteins in periplasmic space are under higher acid stress and are thusmore susceptible to severe damage from acid than the intracellular protein, and these are also more susceptible to severe damage from acid than the intracellular protein. During the acid-resisting process of Gram-negative bacteria, in addition to the intracellular decarboxylase system, the molecular chaperone can also participate in the identification and protection of the space structure of protein as an important "correcting" mechanism. This paper, reviews the current researches on the function, structure, acid-resisting mechanism of variousmolecular chaperones, including HdeA, HdeB, DnaK and GroEL. Finally, the research progress of the acid-resisting mechanism among Gram-negative bacteria is summarized. Through in-depth investigation and analysis of the physiological adaptation strategy of molecular chaperone to the acid stress environment, this study can help to conduct physiological property modification of target strain and improve their viability and tolerance in acid stress environment, which has its important theoretical and practical significance.

Fluorescence imaging can provide valuable information on the expression, distribution, and activity of drug target proteins. Chemical probes are useful small-molecule tools for fluorescence imaging with high structural flexibility and biocompatibility. In this review, we briefly introduce two classes of fluorescent probes for the visualization of drug target proteins. Enzymatically activatable probes make use of the specific enzymatic transformations that generally produce a fluorogenic response upon reacting with target enzymes. Alternatively, specific imaging can be conferred with a ligand that drives the probes to target proteins, where the labeling relies on noncovalent binding, covalent inhibition, or traceless la-beling by ligand-directed chemistry.

Objective@#To carry out an assessment on the public health risk of imported infectious diseases in Jinhua, so as to provide evidence for prevention and control strategies.@*Methods @#Twenty-nine imported infectious diseases were recruited. A risk assessment index system was established by Delphi method and analytic hierarchy process. The actual values of each index of twenty-nine imported infectious diseases were obtained through special investigation,literature review and consultation. The risk scores were calculated by the improved technique for sequencing by approximate ideal solution (TOPSIS) and classified according to median.@*Results @#Among 41 experts recruited, 38 experts were engaged in infectious disease prevention and control, 31 had senior professional titles, and all had worked for more than 10 years. Through three rounds of consultation by Delphi method and analytic hierarchy process, the risk assessment index system (target level) included four items in criteria levels, which were response capacity, public health impact, the possibility of import and local transmission and population vulnerability in a descending order of weight; twenty-seven indicators, with international attention level, levels of medical institutions with confirmed cases, the emergency response capacity of Centers for Disease Control and Prevention (CDCs) , and effective vaccines with wide coverage weighed higher. There were eleven high-risk diseases, high response capacities lay in dengue, malaria, Zika virus disease and Chikungunya fever. The response capacities for sixteen of the eighteen low-risk diseases were insufficient, especially in emergency response capacity of CDCs, laboratory testing capacity and hospital infection control ability.@*Conclusion@#Jinhua has strong capacities to deal with the incidence of dengue fever, malaria, Zika virus disease and Chikungunya fever, compared with other imported infectious diseases.

Pancreatic cancer is well known to be the most deadly malignancy with the worst survival rate of all cancers. High temperature requirement factor A1 (HtrA1) plays an important role in cancer cell proliferation, migration, apoptosis, and differentiation. This study aimed to explore the function of HtrA1 in pancreatic cancer cell growth and its underlying mechanism. We found that the expression of HtrA1 was lower in pancreatic cancer tissue compared to the adjacent normal tissue. Consistently, HtrA1 levels were also decreased in two human pancreatic cancer cell lines, PANC-1 and BXPC-3. Moreover, enforced expression of HtrA1 inhibited cell viability and colony formation of PANC-1 and BXPC-3 cells. Overexpression of HtrA1 promoted apoptosis and suppressed migratory ability of tumor cells. On the contrary, siRNA-mediated knockdown of HtrA1 promoted the growth potential of pancreatic cancer cells. In addition, we found that up-regulation of HtrA1 reduced the expression of Notch-1 in pancreatic cancer cells. On the contrary, knockdown of HtrA1 increased the expression levels of Notch-1. Furthermore, overexpression of Notch-1 abolished the anti-proliferative effect of HtrA1 on pancreatic cancer cells. Taken together, our findings demonstrated that HtrA1 could inhibit pancreatic cancer cell growth via regulating Notch-1 expression, which implied that HtrA1 might be developed as a novel molecular target for pancreatic cancer therapy.

RESUMO

Evidence-based medicine (EBM) requires integration of available ‘best’ clinical evidence,clinician's judgment as well as patient's wishes and expectations.In this paper,we addressed the EBM framework for clinical decision making clinical practice in cardiovascular diseases.Evidence evaluated and interpreted are crucial tools for EBM.EBM has had a profound impact on the treatment of cardiovascular diseases,and confirmed the efficacy of certain controversial interventions.On the other hand,certain widely used conventional strategies for cardiovascular diseases had been proved to be ineffective.Yet,the scientific evidence from China is under resourced to impact the ability to achieve effective actions.We suggest to carry out more high-quality randomized clinical trials to support effective EBM clinical practice.

Objective To evaluate white matter nerve fiber changes of children with binocular ametropic amblyopia by applying the technology of diffusion tensor imaging and the whole brain analysis method of deterministic tractography,and analyze its correlation with visual acuity.Methods Fourteen binocular ametropic amblyopia children was collected from the Second Xiangya Hospital of Central South University as study subject of experimental group,14 cases of normal sight children as the control group.All children were scanned by MRI system,conventional MRI examination,3DTlWI scan were made,then echo-planar sequence scanning was used to obtain diffusion tensor imaging.Quantitative analysis was made to all diffusion tensor imaging using deterministic tractography.Using the experimental group tracts with statistic changes of FA value,volume and tract count as the region of interest(ROI),the correlation analysis with vision for each ROI was performed.Results Isoametropic amblyopia children demonstrated low FA values in the right ventral and dorsal pathway,right optic radiation and corpus callosum compared to control group.There was low volume of fibers in the bilateral ventral and dorsal pathway,the left optic radiation and body of corpus callosum compared to control group.The tract count of right dorsal pathway had reduced compared to control group.Among FA value,tract count and volume,FA value had the maximum regression coefficient with visual acuity,the regression coefficient of tract count and volume was small.The relative correlation coefficient of FA value at right optic radiation,right ventral pathway and body of corpus callosum with vision acuity were 0.486,0.534 and 0.456,respectively,the right ventral pathway had the maximum correlation with visual acuity.Conclusion Isoametropic amblyopia patients shows abnormal structure on bilateral optic radiation,bilateral ventral and dorsal pathway and body of corpus cailosum,these may cause the capability loss of object recognition and spatial position recognition.The FA value of right ventral pathway has the maximum influence on visual acuity.

Objective: To assess fluid-attenuated inversion recovery (FLAIR) vascular hyper-intensity (FVH) and explore its relationship with CT perfusion (CTP) penumbral/infarct core mismatch ratio and diffusion weighted imaging (DWI) final infarct volume in acute ischemic stroke (AIS) patients with middle cerebral artery occlusion (MCAO). Methods: The CTP and MRI images of 38 AIS patients with MCAO were reviewed. The FVH score (longitudinal direction) [FVH score (L)] and FVH score (transverse direction) [FVH score (T)] were quantified on the FLAIR images. The FVH score (L) (range, 0-16) was based on a rostrocaudal extension of FVH and the FVH score (T) (range, 0-3) was based on FVH supply of the occluded MCA territory. The mismatch ratio was calculated from the ratio of the [mean transit time - cerebral blood volume (CBV)] lesion/CBV lesion on the CTP images. The DWI infarct volume was measured on the DWI images. Results: The mismatch ratio was larger for the group of FVH score (L)=7~8 than those of FVH score (L)=5~6 and FVH score (L)=3~4 (p=0.03), whereas the DWI infarct volume was smaller (p=0.04). Similarly, the mismatch ratio of FVH score (T)=2~3 group was larger than FVH score (T)=1 group (p=0.01), whereas the DWI infarct volume was smaller (p=0.02). Both FVH score (L) and FVH score (T) correlated positively with mismatch ratio (P=0.02, P=0.001, respectively), but negatively with DWI infarct volume (P=0.03, P=0.004, respectively). Conclusions: Higher FVH score is associated with larger mismatch ratio and smaller DWI infarct volume in AIS patients with MCAO. FLAIR vascular hyperintensity may represent collateral arterial circulation, and may play a role in protecting the ischemic penumbra.

Purpose It has been demonstrated that hepatocytes uptake of Gd-EOB-DTPA obviously decreased,which would result in decreased liver-to-lesion contrast.Therefore,this study was to evaluate the potential of increasing flip angle to improve the image quality of Gd-EOB-DTPA enhanced MR.Materials and Methods A total of 114 patients with focal liver lesions (FLLs),who underwent Gd-EOB-DTPA-enhanced MR of the liver,were enrolled in this retrospective study.39 patients were noncirrhotic group,36 patients were Child-Pugh A group,23 patients were Child-Pugh B group,and 16 patients were ChildPugh C group.Tl-weighted with fat suppression volume interpolated breath hold examination (VIBE) sequence was acquired before and 5 min,10 min,15 min,20 min after the administration of Gd-EOB-DTPA with both conventional low FA (9 °) and high FA (27 °) protocols separately.Signal to noise ratio (SNR),liver to lesion contrast (LLC),the liver to lesion SI ratio (LLSIR) were calculated and analyzed between low and high FA protocol images in each group and each scan time.Results The LLC and LLSIR on GdEOB-DTPA-enhanced MR images using a high FA (27 °) protocol were significantly higher in comparison with the conventional low FA (9°).In Child B group and Child C group,the LLC and the LLSIR with low FA significantly reduced during 15~20 min after contrast agent injection,however with high FA,the value of LLC and LLSIR tended to be horizontal.Conclusion A high FA protocol in comparison with the conventional 9° FA can obviously improve the image quality,which is reliable for liver lesion depiction on GdEOB-DTPA-enhanced MR images,especially to those with severe liver cirrhosis.

OBJECTIVE@#To explore the 2-[fluorine-18]-fluoro-2-deoxy-D-glucose (¹⁸F-FDG) positron emission tomography/computed tomography (PET-CT) manifestations and functions in patients with primary hyperparathyroidism (pHPT). 
@*METHOD@#The diagnosis of 1 case of double parathyroid adenomas secondary to pPTH was reported and the related literature was reviewed.
@*RESULTS@#PET-CT showed two nodus shadow with slightly lower density compared with muscle, clear boundary and uniform density. Simultaneously, images also showed multiple enhanced FDG uptake of bone lesions in skeletal system. The maximum standard uptake values for ¹⁸F-FDG in the destructive bone lesions were more than 10. 
@*CONCLUSION@#¹⁸F-FDG PET-CT is a well-established imaging technique for the diagnosis in pHPT. It not only can clearly visualize systemic lesions and accurately assess the extent of the disease, but also can accurately localize parathyroid adenoma. After resection of parathyroid adenoma, FDG PET-CT imaging can be useful to see the response to treatment and osteoblastic changes in brown tumor.
